Original abstract
Alpha-synuclein is an intrinsically disordered protein whose aggregation into amyloid fibrils is one of the key pathological mechanisms underlying Parkinson ‘s disease. Recent studies have shown that prior to fibril formation, alpha-synuclein can undergo liquid-liquid phase separation (LLPS), forming protein-rich condensates with locally elevated concentrations. Post-translational modifications can modulate these processes. However, their effect on LLPS remains poorly understood. The aim of this study was to evaluate how point mutations – phosphorylation-mimicking T64D and S87D, and the N-terminal methionine substitution M1Q – affect the propensity of alphasynuclein for LLPS and amyloid aggregation in vitro. Recombinant proteins were expressed in E. coli BL21 Star (DE3) cells using a His6-SUMO fusion tag system and an autoinduction Terrific Broth medium. Proteins were purified by two-step Ni2+-NTA affinity chromatography. Amyloid aggregation kinetics were monitored by ThT fluorescence and OD600 under two conditions: without LLPS (shaking with glass beads) and with LLPS (20 % PEG). The secondary structure of aggregates was assessed by FTIR spectroscopy, and condensate morphology was examined by bright-field microscopy. The aggregation kinetics, both in the absence of LLPS and in the presence of LLPS, showed that the T64D mutant aggregated the fastest, reaches the highest fluorescence and optical density, and formed the most abundant aggregates. M1Q exhibited the weakest and most unstable signal. FTIR confirmed the predominant β-sheet structure in the aggregates of all variants. The results indicate that T64D promotes both LLPS and aggregation, S87D inhibits them, and M1Q inhibits them but with inconsistent results. Alfa-sinukleinas yra nestruktūrinis baltymas, kurio agregacija į amiloidines fibriles yra vienas pagrindinių Parkinsono ligos patologinių mechanizmų. Pastaruoju metu nustatyta, kad prieš susiformuojant fibrilėms alfa-sinukleinas gali pereiti į skysčio-skysčio fazių atsiskyrimo (LLPS) stadiją, kurios metu susidaro baltyminiai kondensatai su padidėjusia lokalia koncentracija. Posttransliacinės modifikacijos gali moduliupoti šiuos procesus, tačiau jų poveikis LLPS yra mažai ištirtas. Šio darbo tikslas – įvertinti, kaip taškinės mutacijos (fosforilinimą imituojančios T64D ir S87D bei N-galinio metionino pakeitimas M1Q) veikia alfa-sinukleino polinkį į LLPS ir amiloidinę agregaciją in vitro. Rekombinantiniai baltymai buvo ekspresuoti E. Coli BL21 (DE3) ląstelėse naudojant His6- SUMO žymens sistemą ir autoinduktyvią Terrific Broth terpę. Baltymai buvo išgryninti dviejų etapų Ni2+-NTA afinine chromatografija su tarpiniu SUMO žymės nukirpimu ir gelfiltracija. Amiloidinės agregacijos kinetika tirta stebint ThT fluorescenciją ir OT600 dvejomis sąlygomis: be LLPS (purtant su su stikliniais rutuliukais) ir su LLPS (20% PEG). Agregatų antrinė struktūra įvertinta FTIR spektroskopija, o kondensatų dydis – šviesaus lauko mikroskopija. Agregacijos kinetika tiek ne LLPS, tiek LLPS sąlygomis parodė, kad T64D mutantas agregavo greičiausiai, pasiekė didžiausią fluorescenciją ir optinį tankį bei sudarė gausiausius kondensatus. M1Q – silpniausią ir nestabiliausią signalą. FTIR patvirtino dominuojančią β-klosčių struktūrą visų variantų agregatuose. Rezultatai rodo, kad T64D – skatina tiek LLPS, tiek agregaciją, S87D – slopina, M1Q – slopina, bet su nepastoviais rezultatais.
