Original abstract
As doenças neurodegenerativas, como a doença de Alzheimer (AD), esclerose múltipla (MS) e doença de Parkinson (PD), são particularmente desafiadoras de diagnosticar, requerendo biomarcadores específicos para cada doença que permitam avaliar o início e a progressão dos seus processos neurodegenerativos. A retina, uma parte integrante do sistema nervoso central que é diretamente observável através de meios óticos, como a tomografia por coerência ótica (OCT), tem vindo a ser proposta como uma fonte para tais biomarcadores. Notavelmente, a tecnologia OCT foi adaptada para a investigação animal, permitindo o estudo do início e progressão destas doenças neurodegenerativas. Para analisar a informação contida nos dados de OCT, os métodos de análise de textura têm vindo a ser destacados como ferramentas promissoras para o estabelecimento de biomarcadores retinianos de neurodegeneração. No presente trabalho, diferentes métricas de textura foram utilizadas para caracterizar a estrutura retiniana no envelhecimento saudável, especificamente como é afetada por fatores como a idade e o sexo biológico. Os resultados obtidos revelam diferenças acentuadas baseadas no sexo, disseminadas por diferentes regiões da retina, e que parecem ser dependentes da idade. Além disso, foram realizados estudos longitudinais num modelo animal (murganho) de AD, cujos resultados sugerem que a textura retiniana pode ser usada para identificar sinais precoces de neurodegeneração na retina. Além disso, estas métricas de textura revelaram diferenças significativas entre o modelo animal de doença e uma população de controlo, em vários pontos do ciclo de vida dos murganhos, sugerindo o seu potencial uso como biomarcadores de progressão da AD. Foi ainda indentificado um paralelismo humano-animal. As diferenças de textura encontradas entre o modelo animal de AD e o grupo de controlo são consistentes com as que foram identificadas entre doentes com diagnóstico de AD e controlos saudáveis (HC). Finalmente, os métodos de análise de textura foram aplicados a dados de OCT obtidos de HC e doentes com AD, MS ou PD, contribuindo para o estabelecimento de potenciais biomarcadores de neuroimagem, não invasivos e de fácil acesso, para estes processos de neurodegeneração. As métricas de textura retiniana em imagens da retina geradas a partir de dados OCT demonstram conter informação útil sobre estes processos neurodegenerativos, podendo inclusivamente ser utilizadas para discriminar entre diferentes doenças neurodegenerativas. Neurodegenerative disorders such as Alzheimer's disease (AD), multiple sclerosis (MS), and Parkinson's disease (PD) are particularly challenging to diagnose, calling for disease-specific biomarkers that allow to assesss the onset and progression of their neurodegenerative processes. The retina, an integral part of the central nervous system that is directly observable through optical means such as optical coherence tomography (OCT), has been proposed as a source for such biomarkers. Notably, the OCT technology has been adapted to animal research, allowing for the study of the onset and progression of these neurodegenerative disorders. To extract the information embedded in OCT data, texture analysis approaches have been highlighted as promising tools for the establishment of retinal biomarkers of neurodegeneration. In the present work, texture metrics were employed to characterise the retinal structure in healthy ageing, specifically how it is affected by factors such as age and biological sex. Results reveal marked sex-based differences, spread over different regions of the retina, which appear to be age-dependent. In addition, longitudinal studies were conducted on mice models of AD, which results suggest retinal texture can be used to detect early signs of neurodegeneration in the mouse retina. These texture metrics also reveal significant differences between AD and control mice at several time-points of the lifespan of the mice, hinting at their potential use as disease progression biomarkers. Furthermore, a human-animal parallel was identified, i.e., texture differences found between AD and control mice are consistent with those identified between AD patients and healthy controls (HC). Finally, OCT data obtained from HC and AD, MS, and PD patients were texture-analysed, contributing towards the establishment of potential non-invasive and easily acessible neuroimaging biomarkers for neurodegeneration. It was shown how retinal texture-based metrics embbed useful information on these neurodegenerative processes, and can even be used to discriminate between neurodegenerative disorders.
