Original abstract

In this thesis, three new fluorosubstituted benzoylthiourea derivatives N-(benzyl(methyl) carbamothioyl)-2-fluorobenzamide (L1), N-(benzyl(methyl)carbamothioyl)-3-fluorobenzamide (L2), and N-(benzyl(methyl)carbamothioyl)-4-fluorobenzamide (L3) were successfully synthesized and characterized using the one-drop synthesis method to develop new therapeutic candidates for the treatment of Parkinson's Disease. The structural confirmation of the compounds was performed by 1H NMR, 13C NMR, FT-IR, UV-Vis, and elemental analysis techniques; furthermore, the precise structure of the L1 compound was elucidated by single-crystal X-ray diffraction. It was determined that the L1 compound crystallized in the monoclinic crystal system and the P21/c space group. Within the scope of theoretical studies, molecular geometry optimization, HOMO-LUMO energy analyses, and thermodynamic parameters were calculated using Density Functional Theory. Hirshfeld surface analysis and energy framework analyses revealed that H···H (%43.1) and dispersion interactions are dominant in the stability of the crystal structure. In biological activity evaluations, it was found that the compounds exhibited dose-dependent antioxidant capacity, with the L2 compound showing the strongest DPPH radical scavenging activity with a 94.7% inhibition rate. In antimicrobial tests, the compounds were observed to be moderately effective against Gram-negative bacteria and highly effective against the yeast Candida albicans (MIC value of 31.25 µg/mL). Finally, the inhibitory potential on the MAO-B enzyme associated with Parkinson's disease was investigated by molecular docking methods, and it was determined that the L1 ligand exhibited a strong interaction with a binding affinity of –8.60 kcal/mol. The results obtained indicate that these newly synthesized derivatives could be potential drug candidates with multifaceted biological activities for the treatment of neurodegenerative diseases. Bu tez çalışmasında, Parkinson hastalığının tedavisine yönelik yeni terapötik adaylar geliştirilmesi amacıyla üç yeni flor sübstitüe benzoiltiyoüre türevi N-(benzil(metil)karbamotiyoil)-2- fluorobenzamid (L1), N-(benzil(metil)karbamotiyoil)-3-fluorobenzamid (L2) ve N-(benzil(metil) karbamotiyoil)-4-fluorobenzamid (L3) tek damla sentez yöntemiyle başarıyla sentezlenmiş ve karakterize edilmiştir. Bileşiklerin yapısal doğrulaması 1H NMR, 13C NMR, FT-IR, UV-Vis ve elementel analiz teknikleriyle yapılmış; ayrıca L1 bileşiğinin kesin yapısı tek kristal X-ışını kırınımı yöntemiyle aydınlatılmıştır. L1 bileşiğinin monoklinik kristal sisteminde ve P21/c uzay grubunda kristallendiği belirlenmiştir. Teorik çalışmalar kapsamında, Yoğunluk Fonksiyonel Teorisi kullanılarak moleküler geometri optimizasyonu, HOMO-LUMO enerji analizleri ve termodinamik parametreler hesaplanmıştır. Hirshfeld yüzey analizi ve enerji çerçeve analizleri, kristal yapının stabilitesinde H···H (%43.1) ve dispersiyon etkileşimlerinin baskın olduğunu ortaya koymuştur. Biyolojik aktivite değerlendirmelerinde, bileşiklerin doza bağlı olarak artan antioksidan kapasite sergilediği, özellikle L2 bileşiğinin %94.7 inhibisyon oranıyla en güçlü DPPH radikal süpürme aktivitesini gösterdiği tespit edilmiştir. Antimikrobiyal testlerde, bileşiklerin Gram-negatif bakterilere karşı orta düzeyde, Candida albicans mantarına karşı ise yüksek düzeyde (31.25 µg/mL MİK değeri) etkili olduğu gözlenmiştir. Son olarak, Parkinson hastalığı ile ilişkili MAO-B enzimi üzerindeki inhibitör potansiyeli moleküler docking yöntemleriyle incelenmiş ve L1 ligandının –8.60 kcal/mol bağlanma afinitesi ile güçlü bir etkileşim sergilediği belirlenmiştir. Elde edilen sonuçlar, sentezlenen bu yeni türevlerin nörodejeneratif hastalıkların tedavisinde çok yönlü biyolojik aktiviteye sahip potansiyel ilaç adayları olabileceğini göstermektedir.
