Original abstract

Neben der Erforschung von Stimulatoren und Aktivatoren der löslichen Guanlyatzyklase (sGC) sind in den letzten Jahren sGC-Hemmstoffe rund um die NO/cGMP-Signaltransduktion relevant geworden. Sie dienen der Erweiterung des Verständnisses des Signalweges und werden sogar als Wirkstoffe zur Behandlung von Migräne, Parkinson oder der Sepsis untersucht.Die vorliegende Arbeit beschäftigt sich mit der Isolierung einer sGC-inhibierenden Komponente aus wässrigem Tabakrauchextrakt sowie der dazugehörigen Methodenfindung. Zudem befasst sich die Arbeit mit der Testung des Tabakrauchextraktes auf isolierter sGC, sowie kultivierten glatten Muskel- und Endothelzellen. Um die grundsätzliche Hemmung der sGC-Aktivität durch Tabakrauchextrakt zu zeigen wurden zwei Methoden zur Aktivitätsbestimmung angewandt und miteinander verglichen. Es zeigte sich sowohl beim klassischen Enzymaktivitätsassay mit radioaktiv markiertem Substrat, als auch bei der indirekten Vermessung über die Pyrophosphatbildung im Zuge des kolorimetrischen Assays eine vergleichbare, signifikante Hemmung des Enzyms.Für die Isolierung des vermuteten Hemmstoffes wurde eine Flüssig-Flüssig-Extraktion des Tabakrauchextraktes angewandt. Um die Extraktion von pH-aktiven Substanzen zu erleichtern, wurden die Extraktion bei unterschiedliche pH-Werte durchgeführt. Die erhaltenen Fraktionen wurden in weiterer Folge in verschiedenen Konzentrationen an der isolierten sGC getestet. Es konnte jedoch keine Fraktion gefunden werden, die eindeutig stärker aktiv war. Tendenziell konnte in der sauren und neutralen Fraktion eine stärkere Hemmung beobachtet werden. Es wird deshalb angenommen, dass ein Stoffgemisch für die beobachtete Hemmung verantwortlich ist. Der wässrige Tabakrauchextrakt wurde in weiterer Folge sowohl auf Endothelzellen als auch auf glatten Muskelzellen aus Schweineaorten getestet. Die Zellen wurden dabei mikroskopisch untersucht und die Proteinexpression von endothelialer Stickstoffmonoxid-Synthase (eNOS), sowie von sGCα1 und sGCβ1 mittels Westernblot überprüft. Die Expression der eNOS blieb in den Endothelzellen weitgehend unverändert, wohingegen die Proteinexpression der sGCα1 und sGCβ1 tendenziell abnahm. Zusammenfassend kommt es durch Tabakrauchextrakt sowohl zur Hemmung der Aktivität der isolierten sGC, als auch tendenziell zur Hemmung der Expression von sGCα1 und sGCβ1 innerhalb der Endothel- und glatten Muskelzellen. Welcher Stoff oder welches Stoffgemisch für diesen Effekt verantwortlich ist, konnte im Rahmen dieser Arbeit nicht geklärt werden. In addition to research on stimulators and activators of soluble guanylyl cyclase (sGC), sGC inhibitors have become significant in research around NO/cGMP signal transduction. They serve to increase the understanding of the signaling pathway and are even being investigated for use as agents in the treatment of migraine, neurological diseases such as Parkinson's and sepsis. This work deals with the isolation of an sGC-inhibiting component from tobacco smoke extract and associated method finding. In addition, the work deals with the testing of the tobacco smoke extract on isolated sGC, as well as cultured smooth muscle and endothelial cells. In order to show the basic inhibition of sGC activity by tobacco smoke extract, two methods for determining activity were used and compared with one another. A comparable, significant inhibition of the enzyme was found both in the classic enzyme activity assay with radiolabeled substrate and in the indirect measurement of pyrophosphate formation in the course of the colorimetric assay.Liquid-liquid extraction of the tobacco smoke extract was used to isolate the putative inhibitor. In order to facilitate the extraction of pH-active substances, the extraction was carried out at different pH values. The fractions obtained were subsequently tested in various concentrations on the isolated sGC. However, no fraction could be found that was significantly more active, although a tendency towards greater inhibition could be observed in the acidic and neutral fractions. It is therefore assumed that a mixture of substances is responsible for the observed inhibition.The aqueous tobacco smoke extract was then tested on both endothelial cells and smooth muscle cells from porcine aorta. The cells were examined microscopically and the protein expression of endothelial nitric oxide synthase (eNOS) and of sGCα1 and sGCβ1 was monitored by Western blotting. The expression of eNOS remained largely unchanged in endothelial cells, whereas the protein expression of sGCα1 and sGCβ1 tended to decrease.In summary, tobacco smoke extract both inhibits isolated sGC activity and tends to inhibit sGCα1 and sGCβ1 expression within endothelial and smooth muscle cells. Which substance or mixture of substances is responsible for this effect could not be clarified during this work.
